On August 26, 2023, at the invitation of Associate Professor Xiao Kai, Professor Chang Lingqian from the School of Chemistry at Beihang University visited our department and gave a report entitled "Single cell diagnostic and therapeutic biochips".


Abstract of the report: The latest cell therapies (such as gene editing and immunotherapy) have brought dawn to the precise diagnosis and treatment of major diseases, but conventional delivery methods have two long-term problems in clinical practice: (1) the cyclic delivery paradigm can easily cause drug retention and toxicity in non target organs, and (2) the larger the molecular weight, the lower the efficiency and controllability of entering target cells. We propose a strategy that fundamentally solves the above problems by designing a new type of micro nano biochip to enhance the manipulation ability of single cells: precise control of the electric field to achieve perforation (nanoelectroporation, NEP) in single cells, electric field driven high speed delivery of macromolecules (chemotherapy drugs, DNA, vaccines, etc.) to cells, achieving ideal delivery efficiency (>99%), cell safety (~100%), and dose controllability (Nature Nanotechnology, 2011). For different applications, high-throughput silicon-based nanopore array chips (JACS 2022: Nano Letters 2016; Small, 2015Small, 2016; Small, 2021; Small, 2022), 3D printed microchannel microneedle microperforation chips (Advanced FunctionalMaterials2021), and flexible nanoelectroporation chips (Nano Letters, 2021) have been designed. Recently, in situ gene transfection of skin cells (Nature Ovarian Injury Repair (Under Review) and breast model Nanotechnology have been achieved, 2017) Single cell research (ln Revision), rapid intestinal repair, etc. (Under Review); Based on this technology, extensions have been made to organ chips, cell chips, and molecular chips respectively.
